Physicochemical characteristics of small bioactive molecules and their role in human lipid synthesis
Low molecular weight drugs and drug-like substances are very important for pharmaceutical research. Taking nature as a starting point nitrogenous molecules have been prepared and are currently the subject of our research. Physicochemical parameters including partition coefficient, pKa and solubility values affects bioavailability and so the effectiveness of active compounds.
To describe drug pharmacokinetic in human body analytical methods like chromatography and UV/Vis spectroscopy will be used. Based on OCED guidelines the methods are to be broadened to physiological conditions.
Another main factor is the effect of small bioactive compounds in human metabolism and pathogenesis. The focus here is on metabolites of arachidonic acid. Potential impact of low molecular weight molecules is monitored in stimulated human whole blood. The inhibition or stimulation of the eicosanoid biosynthesis is determined by a sample work up developed in our group. To quantified lipid mediators HPLC and LC/MS-MS-Analytic is used.
