Entwicklung von kationischen Lipidformulierungen als Nukleinsäure Delivery Systeme

Die Verwendung von Nukleinsäuren als Arzneimittel besitzt ein großes Potential für zukünftige Therapieformen. So bietet die Gentherapie, d.h. das Einbringen von genetischem Material in Zellen mit dem Ziel einen therapeutischen Nutzen zu erlangen, eine aussichtsreiche Strategie für die Behandlung genetisch bedingter Erkrankungen. Weiterhin stellt das Einschleusen von kodierender DNA, mRNA und siRNA auch potentielle Therapieformen für ein breites Spektrum an Erkrankungen wie Krebs, AIDS, neurologische und kardiovaskuläre Erkrankungen.

Für die Durchführung der Gentherapie benötigt man spezielle Vehikelsysteme, welche auch als Vektoren bezeichnet werden. Derzeit dominieren die viralen Vektoren die klinischen Studien. Sie stellen aktuell die effektivsten Vektoren, sind aber mit schwerwiegenden Nachteilen verbunden (onkogenes Potential, Immunogenität, geringe Beladungskapazität). Weiterhin sind die Herstellungskosten der Produktion viraler Vektoren vergleichsweise hoch. Deshalb ist die Weiterentwicklung von nicht-viralen Gentransfersystemen von großem Interesse.

Die Synthese neuer Verbindungen stellt dabei ein wichtiges Werkzeug dar. So wurden von uns in den letzten Jahren 3 neue Klassen von aminofunktionalisierten Lipiden etabliert: verzweigte Fettsäureamide sowie die Malonamide der 1. Und 2. Generation. In systematischen Untersuchungen der DNA-Transfereffizienz der neuen kationischen Lipidformulierungen konnten wir sehr effektive Lipidformulierungen identifizieren.

Ziel unserer Arbeiten ist:

  • die Weiterentwicklung von kationischen Lipidformulierungen.
  • die Aufklärung von Zusammenhängen zwischen physikochemischen Eigenschaften der Lipidformulierungen und ihrer Effektivität.
  • die Untersuchung von zellulären Aufnahmemechanismen um Barrieren zu identifizieren, welche die Gentransfereffizienz reduzieren.
  • Entwicklung von therapeutischen Ansätzen

Dabei kommt eine Vielzahl an Untersuchungsmethoden zum Einsatz:

  • Partikelgrößenbestimmung (DLS, TEM, Cryo-TEM)
  • Zetapotentialmessung
  • kalorimetrische Methoden (DSC, ITC)
  • konfokale Laser-Scanning-Mikroskopie
  • Untersuchungen an der Monoschicht (BAM, IRRAS, Flächen-Druck-Isothermen, Epifluoreszenzmikroskopie)
  • Röntgenbeugungsanalysen (SAXS, WAXS)
  • In-vitro Untersuchungen an verschiedenen Zellkulturmodellen
  • Molekularbiologische Methoden (Gelelektrophorese, PCR)
