Synthesis and physico-chemical characterization of perdeuterated ceramides

The skin is the largest organ of the human and has a variety of different functions, e.g. the regulation of the thermal and the water balance.

Another important role is the barrier function of the stratum corneum. The stratum corneum, the outermost layer of the human skin is composed of keratinocytes and an intercellular lipid matrix. The matrix consists of ceramides, cholesterol and free fatty acids. The ceramides play an important role in the molecular structure of the matrix.

With different kinds of physico-chemical methods, e.g. NMR- and IR-spectroscopy, more detailed information about the matrix structure should be received. This knowledge and the comprehension of the lipid structure are necessary for the development of new potential drug delivery systems.

For detailed studies pure ceramides with a defined chain length are essential. The aim of this work is to synthesize different kinds of perdeuterated ceramids and spingolipids.

The measurements are performed by the research group of Prof. Dr. Neubert (Department of Pharmaceutical Technology and Biopharmaceutics, MLU Halle), Prof. Dr. Huster (Institute for Medical Physics and Biophysics, University Leipzig) and Prof. Dr. Brezesinski (Max-Planck-Institute of Colloids and Interfaces, Potsdam-Golm).
